Depends on your target audience. Obviously anyone who has bought a new computer within the last 4 years or so would be able to display 800x600, whether the screen will be updated fast enough on their machine is another matter.

Yes, I would say so. If you're not targeting pre-G3 macs, all models support 800x600 in 16bit color.

Be a bit careful about requiring exactly any resolution... there'll always be a monitor out there that doesn't support it. For example, I'm pretty sure neither of the cinema displays can do 800x600, though obviously they can do larger :)
